Effect of Extract of Citrus Sunki Peel on Gluconeogenesis in H4IIE Hepatoma Cells |

Abstract |
Peel of Citrus sunki traditionally has a potential anti-inflammatory activity and traditionally has been used to alleviate
respiratory symptoms in Jeju, Korea. However, little is known about the metabolic function of Citrus sunki peel. The present
study investigated whether the extract of the fermented Citrus sunki peel might affect glucose production (gluconeogenesis) in
H4IIE rat hepatoma cells. When H4IIE cells were incubated in glucose-free medium overnight, concentration of glucose in
culture medium were elevated and treatment with ethanol extract of Citrus sunki peel (SE) significantly suppressed the glucose
production. Protein levels of phosphoenolpyruvate carboxykinase (PEPCK) were also decreased by treatment with SE.
Hesperidine and rutin, flavonoids present in SE, however, failed to suppress the glucose production. Addition of hydrogen peroxi
dedecreased the viability of H4IIE cells. SE, hesperidin, rutin protected H4IIE cells against the cytotoxicity induced by hydrogen
peroxide. In summary, SE can suppress glucose production in glucose-deficient conditions and protect cells against oxidative
stress. It should be further studied which compounds are responsible for the gluconeogenic activity of SE. |
