Modulatory function of calcium excitability in astrocytes by NMDA receptor antagonists

Abstract
Astrocytes, the predominant cell type in central nervous system, are electrically non-excitable cells. However, astrocytes display their excitability by increasing intracellular calcium concentration. Several lines of evidence have highlighted the importance of astrocytic calcium excitability in both physiological and pathological conditions. Here, we investigated the effects of dissociated anesthetics: ketamine and MK-801 (dizocilpine) known as NMDA (N-methyl-D-aspartate) receptor antagonists, on calcium excitability in mouse cortical astrocytes. We further compared the effects of Ketamine and MK-801 on astrocytic serotonin (5-HT) receptors. In cultured cortical astrocytes, MK-801 increased intracellular calcium concentration and led to further increases in calcium in the presence of 5-HT. However ketamine had no effect on the calcium response in astrocytes. These results suggest that the calcium concentration in astrocytes is reinforced by MK-801 under the modulation of serotonin receptors.
